What is Fladrafinil?

Fladrafinil, also referred to as CRL 40,941 or Fluorafinil, is a eugeroic compound, discovered by the French pharmacists of Lafon Laboratories in the 1970s. It has been labeled as a “smart drug”, or a nootropic, similar to other popular and highly-researched compounds such as Modafinil and Adrafinil. Similarly to these compounds, On the animals tested Fladrafinil has been shown to have beneficial effects on memory, focus, mood, and energy. Researchers have theorized that the wakefulness benefits of Fladrafinil are likely due to its ability to act on the receptors that inhibit the release of adenosine. Fladrafinil also works to prevent the breakdown of dopamine in the brain. The increased dopamine levels leads to the improved feelings of wakefulness, productivity, and overall focus.

 

Fladrafinil vs Adrafinil

Adrafinil is an interesting compound as it is considered a prodrug, meaning it remains inactive until it is converted into Modafinil in the liver. By metabolizing in the liver, the bioavailability of the compound is increased. Both compounds are found to have similar effects such as increased energy and focus and reduced fatigue. However, the primary difference between Fladrafinil and Adrafinil is their half-life. Adrafinil has a half-life of approximately 1 hour while the half-life of Fladrafinil is around 2-3 hours. With a shorter half-life, Adrafinil has to be taken more frequently which can potentially lead to a higher incidence of side effects. Fladrafinil vs Flmodafinil

Flmodafinil is a compound that is practically identical to the popular nootropic, Modafinil, apart from the addition of two fluoro groups. The addition of the two fluoro groups makes the compound more potent than Modafinil resulting in improved bioavailability. Flmodafinil is similar to Fladrafinil as it works by inhibiting the reuptake of dopamine. Fladrafinil Research

Extensive research regarding the effects of Fladrafinil has yet to be conducted, however, there have been brief animal-based studies supporting claims that the compound can affect cognition and levels of aggression.

Initial studies conducted on mice found that administering Fladrafinil led to an improvement in overall cognition that was not seen in the control group. This is most likely due to the interaction between Fladrafinil and various neurotransmitters in the brain. Additionally, Fladrafinil was not found to affect the sleeping habits of the mice at all, which is important to note considering other studies have found that compounds such as Modafinil and Adrafinil can cause insomnia. Further studies have found that Fladrafinil has anti-aggressive properties. Rats were administered either Fladrafinil or a placebo and it was found that the subjects receiving the active drug exhibited far less aggressive behavior than the subjects in the control group.

Another notable study conducted by Dowling et. Al examined how Fladrafinil and various other compounds were affected by thermal degradation. The technology of Gas-chromatography mass-spectrometry (GC-MS) was utilized for this experiment due to its ability to induce analysis-related ambiguities. The different compounds were exposed to the GC heated injection port that had been dissolved in different solvents. It was found that 1,1,2,2-tetraphenylethane (TPE) was formed during the thermal degradation of both Adrafinil and Modafinil, while a fluorinated analogue of TPE was synthesized during the degradation of Fladrafinil.

Fladrafinil producing the TPE analogue indicates that different degrees of ambiguity could skew data found in forensic analysis. Overall, this study highlights the fact that even though GC-MS normally provides reliable data, researchers have to be cautious and aware of any challenges that could arise during analysis.
