What is J-147?

J-147 is categorized as a phenyl hydrazine and known to be a derivative of the compounds Curcumin and Cyclohexyl-Bisphenol A (CBA). J-147 has strong neuroprotective and neurogenic effects that help to combat degeneration due to aging. Furthermore, different studies primarily conducted on mice have concluded that J-147 shows promise in acting as an effective treatment for Alzheimer’s Disease through improving the function of the mitochondria. Researchers have found that the compound is also likely to improve overall cognitive functioning.

The compound works by binding to ATP synthase in order to stop the overproduction of ATP which can lead to premature aging. It is also able to increase the levels of various neurotransmitters in the brain such as brain derived neurotrophic factor (BDNF) and nerve growth factor (NGF), both of which play a role in preserving memory and improving neuroprotection. By binding to ATP synthase, J-147 is able to increase intracellular calcium, leading to activation of the AMPK in a calcium-dependent protein kinase β-dependent manner. This pathway is directly related to longevity within the brain.

Mice experiencing cognitive decline due to aging were treated with J-147. Results found that it reduced the symptoms of aging throughout the brain. This is due to the regulation of acetyl-coenzyme A, which helps to preserve homeostasis in the mitochondria. Furthermore, regulation and improvement of acetyl-coenzyme A levels leads to the inhibition of acetyl-CoA carboxylase 1. The inhibition of this enzyme tends to lead to memory enhancement and effective neuroprotection.

In terms of Alzheimer’s Disease and overall brain health, J-147 has shown to be efficacious in positively affecting both. J-147 has been shown to improve memory by regulating the Amyloid-β metabolism and reducing the levels of β-secretase. Additionally, the compound has been shown to increase levels of Docosahexaenoic acid (DHA) and Glutamate in the brain, both of which are crucial to the retention and restoration of memory in cases of Alzheimer’s Disease. Studies conducted in mice have shown that J-147 also works to improve permeability and homeostasis of the blood-brain-barrier, vascular function, and long-term potentiation.

Further studies emphasize the ability of J-147 to help increase neuroprotection and improve synaptic plasticity as well as overall brain growth. J-147 promotes synaptic plasticity by regulating the expression of Synaptophysin, a protein that is considered a biomarker for synapse loss and is heavily associated with aging. Brain growth is improved through the compound’s ability to increase levels of BDNF and NGF in the brain. Neuroprotection is directly related to the presence of these to neurotransmitters. BDNF specifically has been shown to increase neuroprotective effects, however, it is not always necessary. For example, various studies have found that in mice with induced Alzheimer’s Disease, J-147 can improve glutathione depletion in the neurons and protect against glucose starvation. Additionally, these studies found that when suffering from Alzheimer’s, Heme Oxygenase 1 (HO1) and 5-Lipoxygenase (5-LOX) have the tendency to recruit immune cells and provoke inflammatory responses in the brain. Cognitive Impairments due to Alzheimer’s Reduced with J-147

There is no current cure for Alzheimer’s Disease, but as it was previously mentioned, J-147 has shown promise in reversing the effects of the condition. Researchers Prior et. Al examined how potent doses of J-147 can affect memory and neuroprotective pathways in the brain of 20-month-old transgenic AD mice. In order to measure results, the researchers performed histology tests, cognitive behavioral assays, Western blotting, and ELISA tests. Additionally, a scopolamine-induced model of memory impairment compared the effects of J-147 to common memory impairment drug, donepezil.

Following the various tests conducted, results found that administration of J-147 can reverse the cognitive impairments of Alzheimer’s even at late stages of the disease. The improvement of memory was found to be tied to the increase in the neurotrophic factors, NGF and BDNF, as well as many of their related proteins that are all crucial to learning and memory. In regards to the scopolamine model comparing J-147 and donepezil, both compounds were effective at restoring short term memory, however, J-147 was more capable of restoring spatial memory as well.
