Characteristics of Uridine Monophosphate

Uridine monophosphate (UMP) is one of the four pyrimidine nucleosides that act as a crucial portion of RNA. RNA is considered a messenger molecule that carries the blueprint for DNA to ribosomes that are responsible for protein synthesis. The presence of uridine is the main difference between RNA and DNA. During the process of translation, the thymine nucleoside base found in DNA turns to the uridine nucleoside base in RNA. Without uridine present, DNA would not be able to activate the mechanism of action for protein synthesis. In addition to being a building block of RNA, uridine assists in developing neurotransmitters and creating new synapses.

Research has found that when administered as a nootropic supplement, UMP can elicit benefits such as enhanced cognition, reduction of age-related decline, and improved mood in animal models. UMP is capable of easily passing the blood-brain-barrier and has proven to have better bioavailability than uridine obtained from food. After serum concentrations of uridine pass the blood-brain-barrier, levels of CDP-choline quickly increase. CDP-choline acts as a source of choline. The compound is unique as it separates in the stomach to cytidine and choline, but after passing the blood-brain-barrier the two particles join back into CDP-choline. CDP-choline is also considered a precursor to phosphatidylcholine (PC), a fatty acid crucial to the synthesis of acetylcholine (ACh). Proper ACh levels in the brain are crucial for cognitive performance and functioning (https://nootropicsexpert.com/cdp-choline/).

Effects of Uridine Monophosphate on Memory

ACh is also crucial to memory and learning as it promotes signal transmission throughout the brain. Various animal-based studies have found that subjects with induced memory loss diseases such as Alzheimer’s or dementia, typically have very low levels of ACh present. Memory is highly dependent on neuroplasticity and synaptic plasticity; ACh is vital to these mechanisms, however, UMP has been shown to influence memory retention and learning due to the role it plays in synapse formation and neuron growth. Researcher Wurtman, et. Al discovered that uridine is involved in the promotion of synapse formation by enhancing neurite growth through the activation of P2Y2 receptors. In addition to synaptogenesis, uridine is indirectly related to neurogenesis. New neurons are found in the phospholipid bilayer, composed of PC. PC is a product of CDP-choline which is significantly increased by uridine.

A team of Alzheimer’s researchers at MIT discovered that a lot of the cognitive decline experienced with Alzheimer’s disease is due to loss of synapses. The researchers decided to administer a “cocktail” of brain-boosting supplements to the rat test subjects. The “cocktail” was composed of DHA (Omega-3), uridine, and choline; the combination of all three was compared to the effects of each compound by itself. Each group showed an increase in dendritic spines, however, the group receiving all three compounds experienced the most improvement in synapse growth and cognitive abilities. Results of the study even reported that the combination group showed signs of synaptic regeneration, which is typically considered a very rare and surprising occurrence. This solidifies the claim that UMP is directly related to synaptogenesis and can promote memory formation while decreasing instances of cognitive delay (http://news.mit.edu/2007/alzheimers-1126).

Effects of Uridine Monophosphate on Mood

In addition to cognitive enhancements, UMP has also been shown to improve mood and combat symptoms of depression by stimulating dopamine in the brain. As it was previously mentioned, the fatty acid, PC is crucial to cellular growth and repair. PC is a derivative of CDP-choline, a compound that dramatically increases when UMP is administered. Researchers Wang et. Al identified that when exogenous uridine levels were increased in rat PC12 cells, intracellular CDP-choline levels elevated due to the synthesis of uridine triphosphate (UTP). This occurrence allowed for uridine to improve neurite growth via nerve growth factor (NGF). Based on their observations, the researchers were able to conduct a study assessing how supplementation with UMP affects dopamine levels in aged rats. Male Fischer rats consumed a control diet or a diet including UMP. The results of the study found that the rats consuming UMP had higher levels of potassium-evoked dopamine release that correlated with dopamine content in the striatum.

The idea of uridine acting as an antidepressant was solidified by the work of Carlezon et. Al. The researchers hypothesized that the antidepressant effects of both Omega-3 and uridine were due to the effects they have on fatty acids in the brain that help facilitate neurotransmission. This is important to mention as neurotransmitters are typically the cause behind cases of depression. The team subjected rats to a forced swim test to mimic symptoms of stress and depression; the quicker they were to become immobile in the water, the more depressed they were. When dosages of uridine or Omega-3 were lowered, the rats experienced more depression. This indicates that the presence of these compounds allow for more efficient dopaminergic neurotransmission (http://www.ncbi.nlm.nih.gov/pubmed/16055952).
