ACE-031 is a peptide that is considered a soluble form of activin type IIB receptors that are linked to the domain of IgG1 Fc. ACE-031 is best known for its ability to increase muscle mass regardless of the type of muscle fibers present. ACE-031 works to block the inhibition of muscle growth. The peptide is known to bind to myostatin and other similar ligands that limit muscle and protein synthesis. Numerous studies show that in cases of muscular dystrophy, muscular degeneration, or muscular injury, myostatin deficiency is actually beneficial to the subject due to myostatin’s ability to inhibit muscle growth. In mice subjects that experienced a myostatin deficiency, hypertrophic and hyperplastic muscle growth increased by 200-300%.

Additionally, studies show that ACE-031 has effectively treated various myopathies such as Duchenne muscular dystrophy (DMD) and amyotrophic lateral sclerosis (ALS) by significantly increasing the muscle mass and body weight of the test subjects in varying animal studies. Effects of ACE-031 on Muscle Mass

A study conducted by Cadena et. Al examined the effects that ACE-031 had on muscle mass in 8-week-old C57BL/6 mice. The mice were treated with either ACE-031 or a vehicle over the course of 28 days. After 28 days, the mice treated with ACE-031 had a 16% increase in mean body weight. Additionally, the researchers measure the muscle gain and the fiber type in the gastrocnemius, soleus, plantaris, and the extensor digitorum longus muscles, the results showed that in the experimental group there was a respective 46, 33, 44, and 26% increase in muscle mass.

The study also examined how ACE-031 affected the types of muscle fibers present. It was found that the mean cross-sectional area of the plantaris, which primarily contains type II muscle fibers, increased by 57%. On the other hand, in the soleus muscle of the treated mice, the mean cross-sectional area saw a 22% increase in type 1 muscle fibers and a 28% increase in type II muscle fibers. However, the fiber type distribution in the soleus of the experimental group remained the same as the fiber type distribution in the control group. The study found that ACE-031 did not precisely differentiate between type I and type II fibers, and cross-sectional areas taken from the experiment group showed that administration of ACE-031 regardless of the fiber type, increased overall muscle growth. ACE-031 and Bone Density

While studies primarily focus on ACE-031 ability to increase muscle mass, it is also shown to help protect bone density following menopause. Treatment with ACE-031 showed that there was an increase in biomarkers relating to bone formation, as well as a simultaneous decrease in the biomarkers associated with reabsorption of bone. Results of the study show that bone mineral density in the lumbar spine increased by 3.4% while the bone density decreased by 1.5% in the control group.
