Please note that RAD-150 (TLB-150) was not developed by Radius Health, Inc. While the official name is TLB-150, it was coined RAD-150 due to its structural similarity to RAD-140, its parent compound.


Overview of RAD 140

The popular SARM RAD 140, or Testolone, goes through the process of esterification to become RAD 150, also known as TLB 150. Testolone is known to have positive effects on neurodegenerative decline caused by insufficient androgen levels in gonadectomized, male rats.  In a study conducted by Jayaramen et. Al, treatment with Testolone was found to have beneficial neuroprotective effects on the brain due to MAPK signaling. The study also came to the conclusion that when combatting cell death, supplementation with Testolone was equally effective as treatment with testosterone (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3959610/). Furthermore, studies showed that when treating juvenile macaques with Testolone, there was a 10% increase in body mass when treated over a 28 day period. The 10% increase in body mass was found to be predominately an increase in lean muscle mass which was confirmed through the use of DEXA scans. 


Process of Esterification

The process of esterification is driven through a substitution reaction. A compound that is considered an ester is frequently derived from a carboxylic acid. Esterification occurs when an alkyl (-O-) group takes the place of the hydroxyl (-OH) group of the carboxylic acid. The substitution reaction takes place when the carboxylic acid is exposed to a strong acid catalyst such as sulfuric acid, and varying levels of heat. Difference Between RAD 140 and RAD 150 

In order to synthesize RAD 150 from Testolone, it has to undergo the process of esterification. During the substitution reaction, a group of benzoate esters is added to Testolone in order to turn the compound into its esterified version, accounting for the main structural difference between the two compounds. By adding the benzoate ester group, the compound now has increased alkalinity, thus indicating that the esterified compound is more stable than Testolone (https://bodyshock.pro/en/products/rad-150-60-caps-3487.html).  


Furthermore, when comparing Testolone to RAD 150, RAD 150 is similar to other esterified SARMs with an increase in half-life and bioavailability. By increasing the half-life of RAD 150, the compound is considered active for a longer period of time than when supplementation with Testolone. Additionally, when bioavailability is increased, the compound is absorbed at a faster rate. It can be concluded that when Testolone undergoes esterification, there are several additional benefits that can be derived from the resulting compound, RAD 150.
