What is Bemethyl?

Bemethyl is a nootropic discovered in the Soviet Union in the 1970s. The compound is considered an alternative to the nootropic Bromantane and is a synthetic actoprotector known for its adaptogenic benefits that allow for efficient responses to external stressors. Actoprotectors are synthetic adaptogens primarily known for their extensive ability to stabilize physiological functioning without excess heat production or oxygen consumption. That being said, animal-based studies have found that actoprotectors are shown to be most effective in subjects with low to medium resistance to extreme conditions. Additional benefits of actoprotectors range from reduced ischemia due to improved protein synthesis, as well as the natural healing and strengthening properties the compound possesses.

Actoprotectors are shown to have stimulatory effects, however, they have different mechanisms than traditional psychostimulants such as modafinil and adrafinil. As it was previously mentioned, actoprotectors do not lead to increased consumption of oxygen or production of heat. The most notable difference between the two groups is that the stimulatory effects of actoprotectors do not involve exhaustive measures as they typically do with psychostimulants.

Bemethyl is known as one of the most potent and highly researched actoprotectors available for research usage. The compound is a member of the benzimidazole family which is what categorizes Bemethyl as a standard actoprotector. Similar to most imidazole compounds, its absorption occurs in the alimentary tube and is enhanced by meals high in carbohydrates. Research has found that following absorption, Bemethyl passes through the blood brain barrier and is metabolized through the liver. Effects of Bemethyl on Physical Performance

The pharmacological effects of Bemethyl are largely due to its structural similarity to the nucleotide bases, guanine and adenine. This allows for increased RNA and protein expression throughout the skeletal muscles and the brain. Originally, Bemethyl was tested on animals in order to see the effects the compound had on physical performance. The compound was shown to increase the rate of ATP production and the rate of gluconeogenesis. This was an important discovery considering gluconeogenesis allows for the production of glucose from sources that are not carbohydrates, for example, lactate, fats, and amino acids. All three play a crucial role in energy production during exercise when carbohydrate-derived glucose storages are depleted.

Research regarding the benefits of Bemethyl have emphasized the compound’s potential to improve physical performance. Various experiments conducted on mice and rats show that administration of Bemethyl leads to increased work capacity and accelerated recovery time under exhaustive loads. Mice treated with Bemethyl exhibited a 33% increase in maximal volume of the work performed and a 60% increase in resistance against tiredness. This led researchers to come to the conclusion that the stimulatory effects of the compound mostly occur during the tiredness phase of physical work (


Biotransformation Pathways Related to Bemethyl

Researchers Belinskaia et. Al investigated how Bemethyl is metabolized in the body of rats in order to enhance their knowledge on the adaptogenic qualities of the compound for further research examinations. Results of the study were measured through the use of liquid chromatography-high-resolution mass spectrometry and in silico examinations. Rats were administered one dose of 330 mg/kg of Bemethyl; urine was then collected and examined for metabolites over the course of 24 hours. After the observation period the urine samples were prepared for analysis by high performance liquid chromatography-tandem mass spectrometry.

The analysis found that there were 9 metabolites in the rat urine with 6 different molecular formulas. The most common metabolite was a benzimidazole-acetylcysteine conjugate which is highly involved in the detoxification of xenobiotics. Xenobiotics are synthetic chemical substances that are considered foreign objects in the body and can cause harm when not regulated. In silico studies were conducted by the BioTransformer and GLORY software programs in order to understand and view the mechanism of the biotransformation pathways of the metabolites.

While these tests cannot find the exact metabolites, chemical formulas, or mechanisms of Bemethyl, they can provide accurate and useful data. This study was highly successful in providing crucial information regarding the biotransformation pathways of the compound.
