GW-0742 is a SARM that belongs to a family of peroxisome proliferator-activated receptors (PPARs) agonists. Studies conducted on mice tend to show that treatment with PPAR agonists are involved in the metabolism of carbohydrates and lipids as well as playing a role in the reduction of inflammation in the gut and regulation of metabolic homeostasis.
Effects of GW-0742 on Insulin and Glucose Levels
In a study conducted by Niu et. Al, researchers were testing the effects GW-0742 has on insulin resistance as well as levels of blood glucose by feeding male Wister rats weighing between 260 and 280, a high fructose diet.
The rats used in this study were fed a 60% fructose-rich diet over a span of two months in order to induce insulin resistance and type 2 diabetes. While given the fructose-rich diet, the blood glucose levels and insulin levels in the rats increased dramatically. The rats were then injected with GW-0742 in order to measure changes in blood glucose levels and insulin sensitivity. After the injection, the study reported that supplementing GW-0742 when the rats were in a metabolically compromised state, levels of insulin resistance and levels of blood glucose considerably decreased (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4610778/).
Effects of GW-0742 on Fuel Utilization
GW-0742 has also been shown to have an effect on fuel utilization throughout the body. A fascinating study conducted by Constantin et. Al measured the effects GW-0742 supplementation has on the fuel pathways.
Over the course of 6 days, male Wister rats were injected with either active GW-0742 or a vehicle at either 5 mg/kg or 100 mg/kg. Over the course of consumption, researchers found that supplementation with GW-0742 caused fuel pathways to shift from using carbohydrates to using lipid oxidation for energy purposes. This was due to increased PDK mRNA as well as increased protein expression (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2277240/).
Effects of GW-0742 on Gut Injury and Inflammation
In addition to the studied effects on insulin resistance, glucose levels, and fuel utilization, Di Paola et. Al conducted a study to determine how GW-0742 affects gut ischemia and reperfusion injury or IRI in the intestines in mice. Researchers started by clamping the celiac trunk and the superior mesenteric artery for approximately 45 minutes. The study showed that by injecting the mice with 0.1 mg/kg of GW-0742 5 minutes prior to removing the clamps and allowing reperfusion there was a reduction in mortality rate, apoptosis, levels of inflammatory cytokines and enzymes, and evidence of gut injury caused by the IRI (https://pubmed.ncbi.nlm.nih.gov/20430778/).
